Butyrate's anti-inflammatory effects in the gut are well documented in human clinical trials.
Key study: A randomized controlled trial of patients with ulcerative colitis found that butyrate enemas (combined with oral mesalazine) significantly improved clinical and endoscopic outcomes compared to mesalazine alone, demonstrating butyrate's direct anti-inflammatory effects on the gut mucosa.

Metabolic health and insulin sensitivity
Gut health and metabolic health are deeply connected. Butyrate has demonstrated effects on blood sugar regulation and insulin sensitivity.
Key study: A randomized, double-blind, placebo-controlled crossover trial found that oral butyrate supplementation (4 g/day for 4 weeks) improved insulin sensitivity in lean men, as measured by hyperinsulinemic-euglycemic clamp. These findings suggest butyrate may help protect against metabolic dysfunction.

GI pain and bloating reduction (especially in women)
A large clinical trial showed particular benefits for women, especially postmenopausal women.
Key study: A 6-week, randomized, double-blind, placebo-controlled trial of 596 participants found that ButyraGen (tributyrin, 200 mg/day) significantly reduced belly pain compared to placebo. Women experienced a 20.3% greater reduction in belly pain and 14.9% decrease in gas and bloating vs. placebo. Postmenopausal women showed the greatest benefit: a 26.4% greater improvement in belly pain scores.

Tolerability and anti-inflammatory markers
Key study: A pilot study of 29 healthy adults taking ButyraGen tributyrin (200 or 400 mg daily for 21 days) showed excellent tolerability (less than 10% reported GI discomfort) and trends toward decreased high-sensitivity C-reactive protein, suggesting systemic anti-inflammatory benefits even at low doses.
