Key study: Researchers at Mayo Clinic and the University of Minnesota screened 10 flavonoids for senolytic activity and found that fisetin was the most potent natural senolytic identified, reducing senescence markers and extending health span and lifespan when administered to aged mice. First published RCT: Joint health and physical function

The first published randomized controlled trial of fisetin as a standalone senolytic showed meaningful results for osteoarthritis.

Key study: In 74 subjects (60% female, mean age 63) with knee osteoarthritis, fisetin was safe with no serious adverse effects over 12+ months. Notable improvements were observed in 4 key physical function tests after 6 months, suggesting that clearing senescent cells in joint tissue slowed cartilage degeneration.

Neuroprotection and cognitive health

Fisetin crosses the blood-brain barrier, making it particularly relevant for protecting brain health with age.

Key study: Research has demonstrated that fisetin activates signaling pathways involved in memory consolidation (ERK/CREB) and has shown neuroprotective properties in multiple human cell studies and clinical investigations, with ongoing trials examining its effects on Alzheimer's disease and age-related cognitive decline.

Ongoing Mayo Clinic human trials

Multiple human trials of fisetin are currently underway, targeting conditions from osteoarthritis to frailty to COVID-19 recovery. The AFFIRM trial (Alleviating Frailty, Inflammation, and Related Measures in Older Adults) is the largest senolytic trial in history.
